A 100 Kb DNA fragment has to be cloned in a host cell. Which vector should be used for this experiment?  A. Plasmid  B. Cosmid  C. BAC  D. Bacteriophage lambda

BAC (Bacterial Artificial Chromosome) is the most suitable vector for cloning a large DNA fragment of 100 Kb. Plasmids have a limited cloning capacity, usually around 10-15 Kb, whereas cosmids can carry larger inserts up to 45 Kb. BACs, on the other hand, have a cloning capacity of up to 300 Kb, making them ideal for cloning large DNA fragments such as the one in this experiment. Bacteriophage lambda can also carry large inserts, but their cloning capacity is limited to around 25-30 Kb, and they are generally more difficult to work with than BACs.
